What it is

Melanotan I (afamelanotide) is the approved variant used clinically as Scenesse for erythropoietic protoporphyria. Unlike Melanotan II it is MC1R-selective, which avoids most of the libido and nausea effects associated with MC4R activation. Pigmentation develops gradually over weeks of subcutaneous loading dosing.

Mechanism

Selective α-MSH analog at the MC1R receptor, stimulating eumelanin synthesis in melanocytes for UV-protective pigmentation.

Generally well tolerated. Monitor moles for change — any melanocortin agonist can stimulate existing nevi.
